Serology
DNA testing (DQ alpha) was performed on blood samples submitted to the Crime Lab. Sex determination was accomplished via analysis at the amelogenin (AMEL) locus. This gene codes for a constituent of human tooth enamel, the protein amelogenin. The AMEL locus is found on the sex chromosomes. In females, identical alleles of 106 base pairs (bps) are found on each X chromosome. In males, slightly different sized alleles occur: a 106bps allele on the X chromosome and a 112bps allele on the Y chromosome. This difference in size allows the determination of the sex of the donor. A sample of DNA obtained from a female will show one band (representing BOTH X alleles) of 106bps. A sample of DNA from a male will show two bands: one at 112bps and one at 106bps. Mixtures of samples will show a variation in band intensity. Theoretically, an equal amount of male and female DNA will show only two AMEL alleles: one at 106bps and one at 112bps. However, the band at 106 will be 3X as intense (2 from female + 1 from male).

DQ Alpha Analysis
Given the nature of the samples, except in the case of known blood samples, mixed samples are possible. When 3 or more alleles or varying intensities of AMEL alleles are observed, a mixture is confirmed. However, from this analysis, it may not be possible to determine the exact number of donors
